Well, a Nuzlocke is a fan-made game mode that has some rules
- You can only catch one Pokemon per Route/Area
- If a Pokemon faints, it's considered dead and can not be used again
- A "dead" Pokemon must be released (or boxed, if you prefer.)
- You give all Pokemon nicknames. This is so you grow more of an attachment to the Pokemon. It wasn't always an official rule, but most people abide by it.
Those are the main ones, but some people have their own rules, adjusted to their play style, which include species clause (if the first Pokemon you find on a route is one you already caught, you get another chance to find a Pokemon), shiny clause (you can catch/use a Pokemon you find if it's shiny, even if it wasn't the first Pokemon on that route), etc.
I am doing this game mode in Pokemon White 2, just because it has a lot more Pokemon, Moves, and Abilities in it.
I am always losing good Pokemon to training. 
It's been against wild encounters both times, too. Not even trainers.
