When you're talking look at his eyes. A straight guy won't pay much attention to how you look and he won't be overly worried about his impression on you. Also if you catch him starring at you when you look away that's usually an easy tell.

It's worked for me on countless occasions. I can usually tell if someone is gay just by having a conversation with them and paying attention to their body language. They don't necessarily have to be feminine.
