In general no it will not stop your computer from getting a virus, that is why you should have virus protection. Ads are generally safe but annoying but not all ads get screened carefully and some malware slips though.
Edit:
I work with a lot of ad companies so for example if I want a banner ad and no full screen ads that is what I expect. Sometimes though with those settings they will allow a full screen ad to slip through and it is really a pain in the ass because they aggregate from so many sources they can't control the ads themselves directly.
