1. two clearly marked singers (ie: [male vocalist 1], [male vocalist 2])
2. the two singers must have different perspectives (ie: one loves ice cream, the other hates ice cream)

Definitely the first. The second is preferred as well, but you could make a duet work even with both voices liking ice cream if that makes sense.
