Switching for newbie(Jeff/Jackie) = no penalty.

Switching Meg for Liz = penalty.

You cannot have an all female/ all male team.

Assuming there's no twist that will randomly force them to leave - yes, they're permanent. I'm guessing this is part of the main twists while the weekly twist will be whatever is going to happen to Da'Vonne and Vanessa.
